RESUMO

Cloud computing alludes to the on-demand availability of personal computer framework resources, primarily information storage and processing power, without the customer's direct personal involvement. Cloud computing has developed dramatically among many organizations due to its benefits such as cost savings, resource pooling, broad network access, and ease of management; nonetheless, security has been a major concern. Researchers have proposed several cryptographic methods to offer cloud data security; however, their execution times are linear and longer. A Security Key 4 Optimization Algorithm (SK4OA) with a non-linear run time is proposed in this paper. The secret key of SK4OA determines the run time rather than the size of the data as such is able to transmit large volumes of data with minimal bandwidth and able to resist security attacks like brute force since its execution timings are unpredictable. A data set from Kaggle was used to determine the algorithm's mean and standard deviation after thirty (30) times of execution. Data sizes of 3KB, 5KB, 8KB, 12KB, and 16 KB were used in this study. There was an empirical analysis done against RC4, Salsa20, and Chacha20 based on encryption time, decryption time, throughput and memory utilization. The analysis showed that SK4OA generated lowest mean non-linear run time of 5.545±2.785 when 16KB of data was executed. Additionally, SK4OA's standard deviation was greater, indicating that the observed data varied far from the mean. However, RC4, Salsa20, and Chacha20 showed smaller standard deviations making them more clustered around the mean resulting in predictable run times.

RESUMO

For amputees, amputation is a devastating experience. Transfemoral amputees require an artificial lower limb prosthesis as a replacement for regaining their gait functions after amputation. Microprocessor-based transfemoral prosthesis has gained significant importance in the last two decades for the rehabilitation of lower limb amputees by assisting them in performing activities of daily living. Commercially available microprocessor-based knee joints have the needed features but are costly, making them beyond the reach of most amputees. The excessive cost of these devices can be attributed to custom sensing and actuating mechanisms, which require significant development cost, making them beyond the reach of most amputees. This research contributes to developing a cost-effective microprocessor-based transfemoral prosthesis by integrating off-the-shelf sensing and actuating mechanisms. Accordingly, a three-level control architecture consisting of top, middle, and low-level controllers was developed for the proposed prosthesis. The top-level controller is responsible for identifying the amputee intent and mode of activity. The mid-level controller determines distinct phases in the activity mode, and the low-level controller was designed to modulate the damping across distinct phases. The developed prosthesis was evaluated on unilateral transfemoral amputees. Since off-the-shelf sensors and actuators are used in i-Inspire, various trials were conducted to evaluate the repeatability of the sensory data. Accordingly, the mean coefficients of correlation for knee angle, force, and inclination were computed at slow and medium walking speeds. The obtained values were, respectively, 0.982 and 0.946 for knee angle, 0.942 and 0.928 for knee force, and 0.825 and 0.758 for knee inclination. These results confirmed that the data are highly correlated with minimum covariance. Accordingly, the sensors provide reliable and repeatable data to the controller for mode detection and intent recognition. Furthermore, the knee angles at self-selected walking speeds were recorded, and it was observed that the i-Inspire Knee maintains a maximum flexion angle between 50° and 60°, which is in accordance with state-of-the-art microprocessor-based transfemoral prosthesis.

RESUMO

BACKGROUND: In recent decades, high-tech prostheses, including microprocessor-controlled knee (MPK), have been developed to improve the functional abilities of lower limb amputees and to reduce gait asymmetry for the prevention of early joint degradation of the intact limb. The aim of this study was to determine the differences in joint moment and power of the intact limb of transfemoral amputees (TFAs) with an MPK compared to healthy individuals in 2 walking speed conditions. METHODS: Twenty-one TFAs with MPK and matched 21 healthy individuals performed a walking task at spontaneous and rapid self-selected speeds. Spatiotemporal gait parameters and intact limb kinetic data were recorded. RESULTS: The hip and knee moments in the frontal plane during rapid walking were not significantly higher than spontaneous walking in TFA group (respectively p = 0.08 and p = 0.48) and were lower than the control group. In the sagittal plane, the hip extensor moment in TFA was higher than the control group in the landing phase (p < 0.001 in both speed conditions). SIGNIFICANCE: The kinetics of the intact limb of active TFAs with an MPK showed a significant reduction at the knee internal abductor moment compared to the control, potentially limiting risk factors for knee osteoarthritis. However, in the sagittal plane, higher hip extensor moments could favor low-back pain appearance. Gait analysis of lower limb amputees should thus be performed to highlight these kinetic adaptations and then help to propose the most relevant rehabilitation and prevention exercises to limit the appearance of early musculoskeletal degeneration.

RESUMO

The Microprocessor complex (MP) is a vital component in the biogenesis of microRNAs (miRNAs) in animals. It plays a crucial role in the biogenesis of microRNAs (miRNAs) in mammals as it cleaves primary miRNAs (pri-miRNAs) to initiate their production. The accurate enzymatic activity of MP is critical to ensuring proper sequencing and expression of miRNAs and their correct cellular functions. RNA elements in pri-miRNAs, including secondary structures and sequencing motifs, RNA editing and modifications, and cofactors, can impact MP cleavage and affect miRNA expression and sequence. To evaluate MP cleavage activity with various RNA substrates under different conditions, we set up an in vitro pri-miRNA cleavage assay. This involves purifying human MP from HEK293E cells, synthesizing pri-miRNAs using in vitro transcription, and performing pri-miRNA cleavage assays using basic laboratory equipment and reagents. These procedures can be performed in various labs and improved for high-throughput analysis of enzymatic activities with thousands of RNA substrates.

RESUMO

PURPOSE: This study aimed to determine the effect of the blue light (BL) filter on the different tasks performed on a laptop on a daily basis. METHODS: Forty subjects were required to perform a 45-minute task on the laptop screen with or without the BL filter on two different days. In the first task, subjects were made to watch a video. In the second task, subjects were made to read a passage from the laptop for 3 minutes, and reading speed was calculated. In the third task, subjects were made to enter the data on the laptop. The time taken to fill in the data was recorded. After the completion of the task, questionnaires were administered. Pre- and post-task accommodative tests were performed. RESULTS: Forty emmetropes with a mean age of 21 ± 2 were recruited. In visual performance, the subjective response showed a significant difference in visual fatigue level with and without the filter. A statistically significant difference was seen in reading speed and data entry during task performance with and without the filter. Pre- and post-task accommodative parameters showed variable responses. CONCLUSION: This study showed that the use of a BL filter improves task performance, but subjectively, people experienced more visual fatigue while using the filter.

RESUMO

INTRODUCTION: With increasing availability of reusable biomedical data - from cohort studies to clinical routine data, data re-users face the problem to manage transferred data according to the heterogeneous data use agreements. While structured metadata is addressed in many contexts including informed consent, contracts are to date still unstructured text documents. In particular within collaborative and active working groups the actual usage agreement's regulations are highly relevant for the daily practice - can I share the data with colleagues from the same university or the same research network, can they be stored on a PHD student's laptop, can I store the data for further approved data usage requests? METHODS: In this article, we inspect and review seven different data usage agreements. We focus on digital data that is copied and transferred to the requester's environment. RESULTS: We identified 24 metadata items in the four main categories data usage, storage, and sharing, as well as publication of results. DISCUSSION: While the topics are largely overlap in the data use agreements, the actual regulations of the topics are diverse. Although we do not explicitly investigate trusted research environments, where data is offered within an analytics platform, we consider them a as subgroup, where most of the practical questions from the data scientist's perspective also arise. CONCLUSION: With a limited set of structured metadata items, data scientists could have information about the data use agreement at hand along with the transferred data in an easily accessible way.

RESUMO

The present-day healthcare system operates on a 4G network, where the data rate needed for many IoT devices is impossible. Also, the latency involved in the network does not support the use of many devices in the network. The 5G-based cellular technology promises an effective healthcare management system with high speed and low latency. The 5G communication technology will replace the 4G technology to satisfy the increasing demand for high data rates. It incorporates higher frequency bands of around 100 MHz using millimetre waves and broadband modulation schemes. It is aimed at providing low latency while supporting real-time machine-to-machine communication. It requires a more significant number of antennas, with an average base station density three times higher than 4G. However, the rise in circuit and processing power for multiple antennas and transceivers deteriorates energy efficiency. Also, the data transmission power for 5G is three times higher than for 4G technology. One of the advanced processors used in today's mobile equipment is NVIDIA Tegra, which has a multicore system on chip (SoC) architecture with two ARM Cortex CPU cores to handle audio, images, and video. The state-of-the-art software coding using JAVA or Python has achieved smooth data transmission from mobile equipment, desktop or laptop through the internet with the support of 5G communication technology. This paper discusses some key areas related to 5G-based healthcare systems such as the architecture, antenna designs, power consumption, file protocols, security, and health implications of 5G networks.

RESUMO

BACKGROUND: Individuals with unilateral transfemoral amputation walk with increased levels of asymmetry, and this is associated with reduced gait efficiency, back pain and overuse of the intact limb. This study investigated the effect of walking with a unilateral absence of loading response knee flexion on the symmetry of anterior-posterior kinetics and centre of mass accelerations. METHODS: A retrospective cohort study design was used, assessing three-dimensional gait data from individuals with unilateral transfemoral amputation (n = 56). The anterior-posterior gait variables analysed included; peak ground reaction forces, impulse, centre of mass acceleration, as well as rate of vertical ground reaction force increase in early stance. With respect to these variables, this study assessed the symmetry between intact and prosthetic limbs, compared intact limbs against a healthy unimpaired control group, and evaluated effect on symmetry of microprocessor controlled knee provision. FINDINGS: Significant between-limb asymmetries were found between intact and prosthetic limbs across all variables (p < 0.0001). Intact limbs showed excessive loading when compared with control group limbs after speed normalisation across all variables (p < 0.0001). No improvement in kinetic symmetry following microprocessor controlled knee provision was found. INTERPRETATION: The gait asymmetries for individuals with transfemoral amputation identified in this study suggest that more should be done by developers to address the resultant overloading of the intact limb, as this is thought to have negative long-term effects. The provision of microprocessor controlled knees did not appear to improve the asymmetries faced by individuals with transfemoral amputation, and clinicians should be aware of this when managing patient expectations.

RESUMO

The study of mouse lung mechanics provides essential insights into the physiological mechanisms of pulmonary disease. Consequently, investigators assemble custom systems comprising infusion-withdrawal syringe pumps and analog pressure sensors to investigate the lung function of these animals. But these systems are expensive and require ongoing regulation, making them challenging to use. Here I introduce LungElast, an open-source, inexpensive, and self-contained instrument that can experimentally determine lung elasticity and volumes even in immature mice. It is assembled using custom 3D printed parts and readily available or easily constructed components. In this device, a microprocessor-controlled stepper motor automatically regulates lung volume by precisely driving a syringe piston whose position is determined using time-of-flight LIDAR technology. The airway pressures associated with the lung volumes are determined using compact sensor-on-chip technology, retrieved in a digital format, and stored by the microcontroller. The instrument software is modular, which eases device testing, calibration, and use. Data are also provided here that specify the accuracy and precision of the elastometer's sensors and volume delivery and demonstrate its use with lung models and mouse pups. This instrument has excellent potential for research and educational work.

RESUMO

Third-generation sequencing technologies are being increasingly used in microbiome research and this has given rise to new challenges in computational microbiome analysis. Oxford Nanopore's MinION is a portable sequencer that streams data that can be basecalled on-the-fly. Here we give an introduction to the MAIRA software, which is designed to analyze MinION sequencing reads from a microbiome sample, as they are produced in real-time, on a laptop. The software processes reads in batches and updates the presented analysis after each batch. There are two analysis steps: First, protein alignments are calculated to determine which genera might be present in a sample. When strong evidence for a genus is found, then, in a second step, a more detailed analysis is performed by aligning the reads against the proteins of all species in the detected genus. The program presents a detailed analysis of species, antibiotic resistance genes, and virulence factors.

RESUMO

BACKGROUND: With progressing technology in the portable computing field, laptops are now integral for work, home and social settings. Different working postures adopted by laptop users impose different loads on the relevant muscles, which can be associated with musculoskeletal discomfort in the various body regions. Some Arabic and Asian cultures adopted postures are not well investigated, particularly for the 20-30 years age-group. OBJECTIVE: This study compared muscle activity in the cervical spine, arm, and wrist among different laptop workstation setups. METHODS: In this cross-sectional study, 23 healthy female university students (age = 24.2±2.28 years, range 20-26 years) performed a standardized 10 minute typing task in four different laptop workstation setups: DESK, SOFA, GROUND sitting with back support, and laptop table (LAP-Tab). Differences between electromyography (EMG) muscle activity recorded in the trapezius (TR), cervical extensors (CE), deltoid (DEL), and wrist extensors (WE) were determined using one-way repeated ANOVA measures with a post-hoc Bonferroni test. RESULTS: Significantly higher muscle activity was observed respectively in the workstations of DESK > LAP-Tab > SOFA > GROUND. Significant differences were found between WE muscle activity and the three other muscle groups (p < 0.001). There was a significant interaction between workstations and muscle activity (F(9,264) = 3.81, p < 0.001, = 0.11), where the WE and DEL muscles showed respectively higher and lower muscle activity in all setups. CONCLUSION: Muscles showed variable activity in different workstations such that the GROUND workstation provided the minimum load, while the DESK workstation showed the maximum load on the measured muscle groups. These findings require further investigation in different cultural and gender specific groups.

RESUMO

miRNA biogenesis is a cellular process that produces mature miRNAs from their primary transcripts, pri-miRNAs, via two RNAse III enzyme complexes: the Drosha-DGCR8 microprocessor complex in the nucleus and the Dicer-TRBP complex in the cytoplasm. Emerging evidence suggests that miRNA biogenesis is tightly regulated by posttranscriptional and posttranslational modifications and aberrant miRNA biogenesis is associated with various human diseases including cancer. DGCR8 has been shown to be modified by SUMOylation. Yet, the SUMO ligase mediating DGCR8 SUMOylation is currently unknown. Here, we report that USP36, a nucleolar ubiquitin-specific protease essential for ribosome biogenesis, is a novel regulator of DGCR8. USP36 interacts with the microprocessor complex and promotes DGCR8 SUMOylation, specifically modified by SUMO2. USP36-mediated SUMOylation does not affect the levels of DGCR8 and the formation of the Drosha-DGCR8 complex, but promotes the binding of DGCR8 to pri-miRNAs. Consistently, abolishing DGCR8 SUMOylation significantly attenuates its binding to pri-miRNAs and knockdown of USP36 attenuates pri-miRNA processing, resulting in marked reduction of tested mature miRNAs. Induced expression of a SUMOylation-defective mutant of DGCR8 inhibits cell proliferation. Together, these results suggest that USP36 plays an important role in regulating miRNA biogenesis by SUMOylating DGCR8. Significance: This study identifies that USP36 mediates DGCR8 SUMOylation by SUMO2 and is critical for miRNA biogenesis. As USP36 is frequently overexpressed in various human cancers, our study suggests that deregulated USP36-miRNA biogenesis pathway may contribute to tumorigenesis.

RESUMO

Human Microprocessor cleaves pri-miRNAs to initiate miRNA biogenesis. The accuracy and efficiency of Microprocessor cleavage ensure appropriate miRNA sequence and expression and thus its proper gene regulation. However, Microprocessor cleaves many pri-miRNAs incorrectly, so it requires assistance from many cofactors. For example, SRSF3 enhances Microprocessor cleavage by interacting with the CNNC motif in pri-miRNAs. However, whether SRSF3 can function with other motifs and/or requires the motifs in a certain secondary structure is unknown. In addition, the function of SRSF7 (a paralog of SRSF3) in miRNA biogenesis still needs to be discovered. Here, we demonstrated that SRSF7 could stimulate Microprocessor cleavage. In addition, by conducting high-throughput pri-miRNA cleavage assays for Microprocessor and SRSF7 or SRSF3, we demonstrated that SRSF7 and SRSF3 function with the CRC and CNNC motifs, adopting certain secondary structures. In addition, SRSF7 and SRSF3 affect the Microprocessor cleavage sites in human cells. Our findings demonstrate the roles of SRSF7 in miRNA biogenesis and provide a comprehensive view of the molecular mechanism of SRSF7 and SRSF3 in enhancing Microprocessor cleavage.

RESUMO

One of the most effective vital signs of health conditions is blood pressure. It has such an impact that changes your state from completely relaxed to extremely unpleasant, which makes the task of blood pressure monitoring a main procedure that almost everyone undergoes whenever there is something wrong or suspicious with his/her health condition. The most popular and accurate ways to measure blood pressure are cuff-based, inconvenient, and pricey, but on the bright side, many experimental studies prove that changes in the color intensities of the RGB channels represent variation in the blood that flows beneath the skin, which is strongly related to blood pressure; hence, we present a novel approach to blood pressure estimation based on the analysis of human face video using hybrid deep learning models. We deeply analyzed proposed approaches and methods to develop combinations of state-of-the-art models that were validated by their testing results on the Vision for Vitals (V4V) dataset compared to the performance of other available proposed models. Additionally, we came up with a new metric to evaluate the performance of our models using Pearson's correlation coefficient between the predicted blood pressure of the subjects and their respiratory rate at each minute, which is provided by our own dataset that includes 60 videos of operators working on personal computers for almost 20 min in each video. Our method provides a cuff-less, fast, and comfortable way to estimate blood pressure with no need for any equipment except the camera of your smartphone.

RESUMO

BACKGROUND: Using a laptop for work is gaining rapid popularity, but there is little evidence of how it influences musculoskeletal disorders (MSDs) and functional characteristics of the neck and shoulder area. OBJECTIVE: This study aimed to compare the prevalence of upper body MSDs and functional characteristics of the neck between office workers using a laptop or desktop computer. METHODS: A total of 110 office workers with a mean age of 41±10 years participated. 45 office workers (73% female) used a laptop and 65 office workers (86% female) used desktop computers. The prevalence of MSDs was recorded using the Nordic Musculoskeletal Questionnaire. Active range of motion (AROM), maximal voluntary isometric contraction (MVC) force, joint position error (JPE), and pain-pressure threshold (PPT) of the neck and shoulder area were measured. RESULTS: Laptop users experienced significantly more MSDs in the right shoulder area on the day of participation (p < 0.001, OR = 4.47), during the previous 7 days (p < 0.01, OR = 3.74), and at 6 months (p < 0.01, OR = 3.57). Laptop users also experienced significantly more MSDs in the left shoulder during the previous 7 days (p < 0.05, OR = 2.44). There were no statistically significant differences in any of the functional characteristics of the neck and shoulder area between the groups. CONCLUSION: Using the laptop computer for office work may pose a higher risk of developing MSDs in the right shoulder area, but might not have long-term effects on the functional characteristics of the neck and shoulder area.

RESUMO

Tetrabromobisphenol A (TBBPA) and hexabromocyclododecane isomers (HBCDs) are widely detected in indoor environments, but the research on the accumulation, contamination, and human exposure of TBBPA and HBCDs in electronic products dust is still limited. It is unclear whether electronic products might pose human health risk via dust ingestion and dermal absorption. In this study, the levels and distributions of TBBPA and HBCDs were investigated in the personal computer (PC) interior dust and PC surface (upper and bottom) wipes. The median concentrations of TBBPA in PC interior dust, upper, and bottom surface wipes were 168.1 ng/g, 13.2 ng/m2, and 15.2 ng/m2, respectively. These levels were generally higher than those of HBCDs, which were 95.2 ng/g, 11.7 ng/m2, and 12.3 ng/m2, respectively. No significant correlations were found among the PC upper and bottom surface wipes, and interior dust, indicating different sources of TBBPA and HBCDs in PC interior and surface dust. The TBBPA and HBCDs in the PC interior dust were mainly released from inner PC materials, while the sources of target compounds on the surface wipes were likely from external environments. The exposure values of two occupational populations (including PC owners and PC repair workers) to TBBPA and HBCDs were measured by PC interior dust and upper surface wipes. The results imply dust ingestion (including hand-to-mouth uptake) is the main contributor of the exposure route to TBBPA and HBCDs for both PC owners and repair workers. Compared to PC owners, PC repair workers showed the greater risk in exposure assessment, which should be paid more attention.

RESUMO

PURPOSE: Speech recognition technology (SRT) is increasingly available and may provide opportunities for individuals with disabilities to participate in necessary activities and meaningful occupations. This inquiry methodically collects and reports on research related to SRT for individuals with disabilities and impairments. MATERIALS AND METHODS: Using the Arksey and O'Malley framework and PRISMA guidelines, five databases were queried and indexes of 11 journals were hand-searched for relevant articles. Articles were included if they were published after 2005, involved SRT, written in English, and participants(s) had a disability. Articles were charted, categorised for level of evidence, and findings were summarised. RESULTS AND CONCLUSIONS: Of the 78 articles that were retrieved, 13 met inclusion criteria and were organised into 4 categories: SRT in primary and secondary education, in post-secondary education, for daily living, and without a specified context. No included articles considered SRT in vocational contexts. It was determined that SRT is a tool that may improve participation and independence of individuals with disabilities in multiple contexts. Most studies reviewed were non-empirical. Opportunities exist for providers to continue to develop a rigorous body of knowledge for the use of SRT to meet educational, vocational and daily living needs.IMPLICATIONS FOR REHABILITATIONSpeech recognition technology (SRT) provides a method of access that is widely available in mainstream technology devices such as personal computers, smartphones and mobile devices, and smart speakers/daily living aids.SRT may improve participation and independence of individuals with disabilities in contexts such as primary and secondary education, post-secondary education, and with independent living tasks. Many studies examining SRT are non-empirical, and there is an opportunity for assistive technology professionals and other providers who use assistive technology to continue to develop a rigorous body of knowledge.

RESUMO

Individuals with lower limb amputation have a high incidence of falls. Above-the-knee amputation and diabetes/vascular disease are both risk factors for falls. Microprocessor knee (MPK) technology may reduce falls in this population. The objective was to determine the association between MPKs and reduced injurious falls. A retrospective analysis of injurious falls within a large, national outcomes database was conducted. Inclusion was limited to adult K3 ambulators with unilateral, transfemoral, or knee disarticulation amputation due to diabetes/vascular disease. There were 744 out of 881 individuals that did not receive an MPK. Results showed that 16.3% of non-MPK users experienced an injurious fall compared to 7.3% of MPK users (p = .007). Not having an MPK resulted in significantly increased odds (unadjusted: OR: 2.47, 95% CI: 1.26-4.83, p = .009; adjusted for confounders: OR: 2.52, 95% CI: 1.28-4.94, p = .007) of incurring an injurious fall over a 6-month period. In conclusion, the current study found use of an MPK strongly associated with reduced injurious falls in a population of patients with amputation due to diabetes/vascular disease. The findings strongly support the use of MPK technology to mitigate fall risk, and in particular injurious falls requiring medical intervention.
